بررسی آستانه تحمل و شناسایی سویه باکتریایی مقاوم به کادمیم در خاک های پذیرنده پساب

چکیده
زمينه و هدف: جذب زیستی فلزات سنگین یکی از روشهای نوین جهت پاکسازی محیط میباشد. ورود فلزات سنگین از صنایع مختلف به محیط باعث تغییر ساختار جمعیتی و بروز ساز و کار مقاومتی در میکروارگانیسمها میگردد. هدف این تحقیق، تعیین آستانه مقاومت باکتریها به کادمیم، شناسایی باکتریهای بومی جهت حذف این فلز از خاک و دستیابی به کارایی بیشتر در تصفیه بیولوژیکی پسابهای صنعتی بوده است.
مواد و روشها: دراین تحقیق نمونه برداری از خاک سه شهرک صنعتی همدان انجام گرفته و پس از اندازه گیری غلظت کادمیم در خاک، از نمونهها کشت باکتریایی تهیه شده است. پس از غربالگری، جهت تعیین آستانه تحمل باکتریها، حداقل غلظت ممانعت کننده رشد و حداقل دوز کشنده باکتری مشخص گردید. سپس میزان جذب سویهها مطالعه شد و در نهایت باکتری مقاوم شناسایی شد.
يافتهها: از کشت اولیه با میانگین غلظت 78/0 ، 500 و750 mg/L فلز کادمیم به ترتیب تعداد 42 ،4 و 4 سویه باکتریایی شناسایی گردیدند. بررسی حداقل غلظت ممانعت کننده رشد 4 سویه نشان داد که بالاترین مقدار مربوط به سویههای TCd2 و TCd4با غلظت mg/L 1250 بوده است، ولی نتایج جذب نشان داد که بیشترین جذب توسط سویه TCd2 با 2/30% بوده است که این سویه از خانواده انتروباکتریاسه و متعلق به جنس انتروباکتراست.
نتيجه گيري: باکتری بومی شناسایی شده قابلیت تحمل و جذب غلظت بالایی از فلز کادمیم را دارا میباشد که میتوان از آن در تصفیه زیستی فاضلابها و یا به عنوان بیوفیلتر در صنایع تولید کننده این فلز استفاده نمود.
